Overview

This is a combination regimen of three investigational immunotherapy agents: avelumab, CMP-001, and PF-04518600. - **Avelumab** is a fully human IgG1 monoclonal antibody that targets programmed death-ligand 1 (PD-L1), thereby blocking the interaction between PD-L1 and the PD-1 receptor on T cells. This results in immune activation and anti-tumor effects. - **CMP-001** is a CpG-A oligodeoxynucleotide packaged in a virus-like particle, acting as a Toll-like receptor 9 (TLR9) agonist. It is designed to activate plasmacytoid dendritic cells and enhance anti-tumor immune responses through type I interferon production. - **PF-04518600** is a humanized IgG2 monoclonal antibody agonist of OX40 (also known as CD134 or TNFRSF4), a costimulatory receptor primarily expressed on activated T cells. Agonism of OX40 promotes T cell proliferation, survival, and cytokine production, augmenting anti-tumor immunity. This combination is under clinical investigation primarily in advanced and metastatic cancers, including triple-negative breast cancer, leveraging complementary mechanisms in immune checkpoint inhibition (avelumab), innate immune activation (CMP-001), and T cell co-stimulation (PF-04518600)[8].
