Overview

Avelumab + lenvatinib is a combination therapy consisting of avelumab, a fully human IgG1 monoclonal antibody that targets programmed death-ligand 1 (PD-L1), and lenvatinib, an oral small molecule multiple receptor tyrosine kinase (RTK) inhibitor. **Avelumab** inhibits the interaction between PD-L1 and PD-1, thus preventing immune evasion by tumor cells and activating T cell-mediated antitumor responses, while also enabling antitumor activity through natural killer (NK) cell-mediated antibody-dependent cellular cytotoxicity (ADCC). **Lenvatinib** targets and inhibits the kinase activities of several RTKs implicated in tumor growth and angiogenesis, including vascular endothelial growth factor receptors VEGFR1, VEGFR2, VEGFR3, fibroblast growth factor receptors FGFR1, FGFR2, FGFR3, FGFR4, platelet-derived growth factor receptor alpha (PDGFRα), KIT, and RET. This combination is under investigation in advanced solid tumors, including metastatic renal cell carcinoma and other cancers, generally aiming to enhance both immune-mediated and anti-angiogenic antineoplastic effects[1][2][5][6].
