Avidin-biotin-dota-90y is a radiopharmaceutical complex used in pretargeted radioimmunotherapy. It consists of the radioactive isotope yttrium-90 (a beta emitter) chelated by DOTA (1,4,7,10-tetraazacyclododecane-N,N',N'',N'''-tetraacetic acid), which is covalently linked to biotin. This biotinylated complex binds with high affinity to avidin or streptavidin that has been previously localized at the tumor site via a monoclonal antibody conjugate. The strong avidin–biotin interaction allows for highly specific delivery of radiation to tumor cells while minimizing systemic exposure. The drug has been studied in both animal models and clinical trials for various solid tumors including breast cancer and non-muscle invasive bladder cancer[1][3][5][7][10]. Its mechanism relies on delivering targeted beta radiation to destroy residual or microscopic disease after surgical excision or biopsy.
