AvidinOX is an oxidized form of the glycoprotein avidin that covalently binds to tissue proteins upon local administration, creating a stable anchoring site for biotinylated therapeutics. This property enables targeted delivery of radiolabeled or therapeutic biotin-conjugated agents directly to specific tissues or tumor sites. The primary mechanism involves using AvidinOX as a pretargeting platform for radionuclide therapy—after local injection into tumors, it allows subsequent systemic administration of radioactive or therapeutic biotin conjugates that bind specifically to the anchored AvidinOX in the target tissue. This approach has been investigated primarily for inoperable tumors and metastatic cancers (e.g., head and neck cancer, colorectal cancer liver metastases), aiming to enhance local efficacy while reducing systemic toxicity. Preclinical studies have also shown its utility in combination with biotinylated monoclonal antibodies (such as trastuzumab and pertuzumab) for enhanced anti-tumor effects[2][3][4][5][6].
