**Avidocin-CD291.2** is an engineered R-type bacteriocin (diffocin) developed by AvidBiotics (now Pylum Biosciences) as a precision antibacterial agent specifically targeting **Clostridioides difficile** (C. difficile) ribotype 027 strains and select other ribotypes (001, 015, 087). It binds to the surface layer protein A (SlpA) on target bacteria, delivering bactericidal activity via a modified tail fiber that disrupts cell integrity, killing vegetative cells without affecting spores or the gut microbiota. Preclinical mouse studies demonstrated oral stability through the GI tract, prevention of C. difficile colonization and spore transmission, no disruption to colonization resistance against C. difficile or vancomycin-resistant Enterococcus, and no detectable microbiome alterations, positioning it for prophylaxis against C. difficile infection (CDI) in high-risk patients.[1][2][4][9]
