Overview

Avizakimab (BOS161721) is a fully humanized immunoglobulin G1 (IgG1) triple mutation monoclonal antibody designed to selectively bind and neutralize interleukin-21 (IL-21). IL-21 is a cytokine implicated in the pathogenesis of systemic lupus erythematosus (SLE) and other autoimmune diseases by promoting inflammatory responses through its action on multiple immune cell types. Avizakimab inhibits T-cell dependent antigen responses of B-cells and reverses IL-21-induced gene expression changes associated with tolerance induction and T-cell exhaustion. The drug was developed for subcutaneous or intravenous administration in adults with moderately to severely active SLE. It has demonstrated good safety and tolerability in phase 1 studies in healthy volunteers as well as phase 2 studies in SLE patients[2][3][5][7].
