Overview

AVL-101 is an orally bioavailable, small molecule inhibitor of Bruton's Tyrosine Kinase (BTK) developed by Avila Therapeutics. It utilizes a covalent bonding mechanism to achieve potent and selective inhibition of BTK, a key signaling protein in B-cell development and activation. By irreversibly binding to the target, AVL-101 aims to "silence" the protein, providing a prolonged duration of action. It was primarily investigated for the treatment of B-cell hematological malignancies, such as non-Hodgkin's lymphoma (NHL) and B-cell chronic lymphocytic leukemia (B-CLL).
