AVL-3288 is a first-in-class, orally available, small molecule that acts as a selective positive allosteric modulator (PAM) of the alpha7 nicotinic acetylcholine receptor (α7 nAChR). Unlike direct agonists, it enhances receptor activity only in the presence of the endogenous ligand acetylcholine and does not bind to or activate α7 nAChRs via the orthosteric site. This mechanism is thought to reduce the risk of receptor desensitization. The drug has been investigated for its potential neurocognitive benefits in central nervous system disorders such as schizophrenia and cognitive dysfunction. Preclinical studies have shown efficacy in models of cognitive impairment, and early clinical trials have evaluated safety and preliminary effects on cognition in both healthy volunteers and patients with schizophrenia or schizoaffective disorder[2][5][9].
