Drug intelligence / Profile preview
AVLX-144 is a dimeric peptide-like neuroprotectant developed by Avilex Pharma for the treatment of acute neurological conditions such as acute ischemic stroke and subarachnoid hemorrhage. It acts as a high-affinity inhibitor of postsynaptic density protein 95 (PSD-95), specifically binding to both PDZ domains of PSD-95 simultaneously. This mechanism disrupts the interaction between NMDA receptors and neuronal nitric oxide synthase (nNOS), which is implicated in excitotoxic neuronal death following acute brain injury. The design of AVLX-144 provides enhanced stability, high affinity for its target, and improved neuroprotective properties in vivo. The drug has completed Phase 1 clinical trials and is positioned as a best-in-class neuroprotectant for both orphan and broad indications within neurology[1][2][3][4][5][6].
