Overview

AVN-322 is a potent and highly selective small molecule antagonist of the 5-hydroxytryptamine subtype 6 (5-HT6) receptor, developed by Avineuro Pharmaceuticals in collaboration with AllaChem and ChemDiv. It is designed for oral administration to treat cognitive dysfunction associated with neurodegenerative and neuropsychiatric disorders, such as Alzheimer's disease and schizophrenia. AVN-322 has demonstrated high binding affinity (Ki = 389 pM) and functional potency in blocking the 5-HT6 receptor, with preclinical studies showing its ability to reverse cognitive deficits in animal models. The drug completed Phase I clinical trials in 2010, where it was found to be well-tolerated with favorable pharmacokinetic properties, including significant blood-brain barrier penetration. As of the latest pipeline information, it is positioned for Phase 2 development.
