Overview

AVO-101 is an experimental in vivo CAR-T cell therapy being developed by AAVivo for the treatment of relapsed or refractory multiple myeloma. The therapy utilizes a proprietary adeno-associated virus (AAV) vector, specifically the AAVTCeT capsid, to deliver a chimeric antigen receptor (CAR) transgene directly into a patient's T cells in vivo. This transgene encodes a CAR designed to recognize and bind to B-cell maturation antigen (BCMA), which is highly expressed on the surface of malignant plasma cells in multiple myeloma. By engineering T cells within the patient's body, AVO-101 aims to circumvent the logistical and manufacturing complexities associated with traditional ex vivo CAR-T therapies. As of early 2024, the program is in the preclinical stage, focusing on lead optimization and functional validation.
