Avocatin B is a bioactive lipid compound derived from avocado fruit. It acts as a potent inhibitor of fatty acid oxidation (FAO), primarily by targeting mitochondrial enzymes such as very long-chain acyl-CoA dehydrogenase (VLCAD) and carnitine palmitoyltransferase 1 (CPT1). By inhibiting FAO, avocatin B increases glucose oxidation, reduces mitochondrial reactive oxygen species, and improves insulin sensitivity. In preclinical models, it has demonstrated selective cytotoxicity against acute myeloid leukemia (AML) stem cells while sparing normal hematopoietic stem cells. Its mechanism involves the induction of ROS-dependent apoptosis in cancer cells through decreased NADPH levels and disruption of mitochondrial metabolism. Avocatin B has also shown promise in improving metabolic dysfunction associated with obesity and type 2 diabetes by enhancing glucose tolerance and insulin responsiveness. Human clinical trials have indicated that oral supplementation with avocatin B is well-tolerated at doses up to 200 mg per day[1][2][3][4][5][7].
