AVT-02 UE ointment is a topical therapeutic candidate developed by Avontec GmbH for the treatment of mild to moderate atopic dermatitis. It consists of a 2% concentration of AVT-02, a double-stranded decoy oligonucleotide, formulated in an emulsifying ointment (Unguentum Emulsificans). AVT-02 is designed to simultaneously target and inhibit two key transcription factors, Nuclear factor NF-kappa-B (NF-kappaB) and Transcription factor AP-1 (AP-1), which play central roles in the regulation of pro-inflammatory cytokine expression. By acting as a molecular sink, the decoy oligonucleotide prevents these transcription factors from binding to their cognate sequences in the promoter regions of target genes, thereby suppressing the inflammatory cascade in the skin. The drug was evaluated in a Phase IIa multicenter, randomized, double-blind, placebo-controlled trial using an intraindividual comparison design to assess its efficacy, safety, and tolerability.
