Overview

AVX901 is an investigational cancer vaccine designed to treat cancers that overexpress the human epidermal growth factor receptor 2 (HER2/ERBB2). It is based on virus-like replicon particles (VRPs) derived from an alphaviral vector encoding the extracellular domain and transmembrane regions of HER2. The vaccine uses a modified Venezuelan equine encephalitis virus that cannot cause active infection but delivers genetic material to host cells to express large amounts of the HER2 protein. This stimulates both cellular and humoral immune responses against tumor cells expressing HER2 by activating cytotoxic T lymphocytes and memory CD8+ T cells. Preclinical studies have shown reduced tumor growth in animal models. In clinical trials for breast cancer—including metastatic and advanced cases—AVX901 has demonstrated safety with no dose-limiting toxicities reported so far[1][3][5][6][7].
