Overview

AVZO-021 + fulvestrant is an investigational combination therapy currently in Phase 1/2 clinical trials for advanced solid tumors, specifically targeting hormone receptor-positive, human epidermal growth factor receptor 2-negative (HR+/HER2-) breast cancer and other cyclin E1 protein (CCNE1)-amplified malignancies. AVZO-021 is a novel, orally available, potent and selective cyclin-dependent kinase 2 (CDK2) inhibitor designed to disrupt cell cycle progression by inhibiting the CDK2 enzyme. Fulvestrant is a selective estrogen receptor antagonist that blocks and degrades estrogen receptors, impeding hormone-driven tumor growth. The combination is being studied in the post-CDK4/6 inhibitor setting to address tumors resistant to prior CDK4/6 inhibitor or endocrine therapy, with the aim of enhancing antitumor activity by dual inhibition of cell cycle progression and hormone signaling pathways[3][4][5].
