AVZO-023 + letrozole is an investigational combination therapy under clinical evaluation for advanced or metastatic hormone receptor-positive, human epidermal growth factor receptor 2-negative (HR+/HER2-) breast cancer and select advanced solid tumors. AVZO-023 is a first-in-class, potent, and selective small molecule inhibitor of cyclin-dependent kinase 4 (CDK4), developed to block cell cycle progression in cancer cells with CDK4 or CCND1 amplification. Letrozole is a well-established oral nonsteroidal aromatase inhibitor used as endocrine therapy to reduce estrogen production, thus depriving hormone-dependent breast cancer cells of growth signals. The combination aims to improve efficacy by dual targeting of CDK4-driven cell proliferation and estrogen-driven tumor growth. Clinical studies are testing AVZO-023 as monotherapy and in combination with established endocrine therapies, including letrozole, for patients who have not responded to standard treatments.
