Awake-102 is a preclinical-stage microbial immunotherapy being developed by Matter Bio for the treatment of refractory ovarian cancer. The drug utilizes the LM-LLO-TT platform, which consists of a live-attenuated *Listeria monocytogenes* (LM) vector engineered to express and secrete a fusion protein of listeriolysin O (LLO) and a tetanus toxoid (TT) recall antigen. Upon administration, the *Listeria* vector selectively localizes within the immunosuppressive tumor microenvironment. The delivery of the tetanus antigen is designed to recruit and reactivate pre-existing memory T cells—originally generated through routine childhood tetanus vaccinations—to attack the tumor cells. This approach aims to convert immunologically "cold" tumors into "hot" tumors by bypassing the need for the de novo priming of T cells against neoantigens.
