Awake-103 is an immunotherapy candidate in the discovery stage, developed by Matter Bio for the treatment of glioblastoma. It utilizes the LM-LLO-TT platform, which employs a live-attenuated *Listeria monocytogenes* (LM) vector to deliver a tetanus toxoid (TT) recall antigen fused to listeriolysin O (LLO). The therapy is designed to localize within the tumor microenvironment and present the tetanus antigen, thereby recruiting and activating pre-existing memory T cells—formed during prior routine tetanus vaccinations—to attack the tumor. This strategy aims to overcome the immunosuppressive nature of the glioblastoma environment by leveraging a potent, pre-existing immune memory rather than relying solely on the identification and priming of novel tumor-specific antigens.
