AWT020 is a bifunctional fusion protein immunotherapy developed by Anwita Biosciences. It consists of a humanized anti-PD-1 nanobody fused to an engineered interleukin-2 mutein (IL-2c). AWT020 is designed to combine PD-1 pathway blockade with selective IL-2 signaling, enhancing anti-tumor immune responses while minimizing systemic toxicity. It targets PD-1-expressing tumor-infiltrating lymphocytes (TILs), delivering the IL-2c payload specifically to these cells and promoting their expansion and activation within the tumor microenvironment. The IL-2 mutein component has reduced affinity for the IL-2 receptor alpha subunit, ensuring that its activity is largely restricted to TILs expressing PD-1, which helps avoid widespread immune activation and associated toxicities. It is being evaluated in Phase 1 clinical trials for patients with advanced cancers who have failed standard therapies.
