AWT030 is a first-in-class bi-functional interleukin-21 (IL-21) fusion protein designed to selectively activate tumor-infiltrating CD8+ T cells and suppress regulatory T cells (Tregs) within the tumor microenvironment (TME). Developed by Anwita Biosciences, the molecule comprises a stability- and potency-optimized IL-21 mutein fused to an engineered functional domain that targets T cells. This design aims to enhance anti-tumor activity, tolerability, and half-life compared to wild-type IL-21, while avoiding the suppressive effects of IL-21 on dendritic cells. In preclinical models of breast cancer, fibrosarcoma, and colon cancer, AWT030 has demonstrated powerful synergistic effects when combined with anti-PD-1 therapy, leading to complete tumor regression without noticeable toxicity.
