AX-101 (beta alethine) is a small molecule immunostimulatory agent being developed by Aximmune for the treatment of various cancers, including lymphoma and melanoma. The drug is designed to enhance the immune system's anti-tumor response by promoting lymphocyte proliferation, inducing a Th1 immune profile, increasing T-cell cytotoxicity, and improving the M1/M2 macrophage ratio. AX-101 is particularly noted for its synergistic potential when combined with anti-PD-1 immune checkpoint inhibitors such as pembrolizumab, nivolumab, and cemiplimab. While the exact molecular target of AX-101 has not been fully elucidated, it has demonstrated potent anti-tumor activity in preclinical models of bladder, colorectal, and skin cancers. Historically, the drug was evaluated in Phase 1/2 clinical trials as a monotherapy for follicular lymphoma and myeloma, but current development efforts are focused on its application in combination with checkpoint blockade.
