Overview

AX-1412 is an investigational RNA-editing therapy developed by ProQR Therapeutics. It is designed to introduce a protective variant (p.Asn352Ser) into the B4GALT1 gene, which encodes beta-1,4-galactosyltransferase 1. Human genetic studies have shown that this specific variant is associated with a significantly lower risk of developing cardiovascular diseases (CVDs), including coronary artery disease. The mechanism involves hypo-galactosylation of apolipoprotein B100 and fibrinogen—two independent drivers of CVD risk—resulting in reduced levels of LDL cholesterol and fibrinogen, thereby lowering residual cardiovascular risk. AX-1412 utilizes ProQR’s Axiomer RNA editing platform, which leverages endogenous ADAR enzymes for precise A-to-I (adenosine-to-inosine) editing at the mRNA level. This approach offers pleiotropic cardiovascular protection and represents a novel therapeutic strategy distinct from traditional knockdown or inhibition technologies[1][3][5][7].
