AX-5006 is a small molecule, gut-restricted amyloid inhibitor developed as a potential disease-modifying and symptomatic treatment for Parkinson’s disease. Its mechanism of action involves inhibiting the aggregation and expression of CsgA protein, a bacterial protein critical to the formation of amyloid sheets by gut bacteria. By reducing bacterial amyloid aggregation in the gut, AX-5006 aims to alleviate gastrointestinal symptoms (such as constipation) and slow or modify the progression of Parkinson’s disease. Preclinical studies have shown that AX-5006 improves motor symptoms and reduces alpha-synuclein aggregation in animal models. The drug is designed to act primarily within the gastrointestinal tract, resulting in high fecal concentrations with minimal systemic exposure, which may contribute to a favorable safety profile. IND-enabling studies are ongoing with plans for clinical trials[1][2][3][4][5][6][7].
