AX-TCRT-001 is an autologous T-cell receptor (TCR) engineered T-cell therapy designed to target the NY-ESO-1 (New York esophageal squamous cell carcinoma 1) antigen. Developed by Axe-Bio, the therapy involves the genetic modification of a patient's own T cells to express a high-affinity TCR specific for NY-ESO-1 peptides presented in the context of HLA-A*02:01. NY-ESO-1 is a cancer-testis antigen frequently overexpressed in various solid tumors, including synovial sarcoma, myxoid/round cell liposarcoma, and melanoma, while having limited expression in normal tissues. By redirecting T cells to recognize and eliminate NY-ESO-1-positive cancer cells, AX-TCRT-001 aims to provide a potent and specific anti-tumor response. The therapy is currently being evaluated in clinical trials for patients with NY-ESO-1 positive advanced solid tumors.
