AXA-042

AXA-042 is a fully synthetic, systemically administered Toll-like receptor 2/6 (TLR2/6) agonist developed by Axelia Oncology, an Australian biopharmaceutical company spun out of Ena Respiratory. Functioning as an innate immune modulator, AXA-042 engages the TLR2/6 heterodimer to drive anti-tumor responses, a process shown in preclinical models to be macrophage-dependent. The drug is a pegylated small molecule (often characterized as a synthetic lipopeptide) designed to enhance systemic activity and synergize with checkpoint inhibitors. It is currently being evaluated in a Phase 1 clinical trial for patients with advanced solid tumors, where it is administered as a monotherapy and in combination with the PD-1 inhibitor cemiplimab (Libtayo). The technology is based on research from the Peter Doherty Institute for Infection and Immunity at the University of Melbourne.
