Axicabtagene ciloleucel is a CD19-directed genetically modified autologous T cell immunotherapy, classified as a CAR-T cell therapy. It is prepared by collecting a patient's own T cells, genetically engineering them to express an anti-CD19 chimeric antigen receptor (CAR), and then infusing them back into the patient. These engineered T cells recognize and kill CD19-positive B-cell lymphoma cells. The drug is primarily indicated for adults with relapsed or refractory large B-cell lymphomas—including diffuse large B-cell lymphoma (DLBCL), primary mediastinal large B-cell lymphoma, high-grade B-cell lymphoma, DLBCL arising from follicular lymphoma—and for relapsed or refractory follicular lymphoma after multiple lines of systemic therapy. Axicabtagene ciloleucel was developed by Kite Pharma and represents one of the first FDA-approved CAR-T therapies[3][4][5][6][8].
