Axitinib + bevacizumab is an investigational combination therapy consisting of two targeted antiangiogenic agents. Axitinib is an oral, potent, and selective small molecule inhibitor of vascular endothelial growth factor receptors 1, 2, and 3 (VEGFR1/2/3), while bevacizumab is a recombinant humanized monoclonal antibody that binds to vascular endothelial growth factor A (VEGF-A), preventing its interaction with VEGF receptors. Both drugs disrupt tumor angiogenesis but act at different points in the VEGF pathway—axitinib blocks receptor signaling intracellularly, whereas bevacizumab neutralizes the ligand extracellularly. This combination has been studied in early-phase clinical trials for advanced solid tumors and gastrointestinal cancers to assess safety and pharmacokinetics; it was generally well tolerated at lower doses of bevacizumab but showed increased toxicity at higher doses[1][3][4]. The combination aims to enhance anti-tumor efficacy by dual inhibition of the VEGF pathway.
