Overview

Axitinib + capecitabine is a combination of two oral small molecule drugs used in oncology clinical trials for advanced solid tumors. Axitinib is a potent and selective second-generation tyrosine kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR-1, VEGFR-2, VEGFR-3), thereby inhibiting angiogenesis, tumor growth, and metastasis. Capecitabine is an orally administered prodrug of 5-fluorouracil (5-FU), which acts as an antimetabolite by interfering with DNA synthesis through inhibition of thymidylate synthase and incorporation into DNA and RNA. The combination has been studied in phase I trials for safety, tolerability, pharmacokinetics, and preliminary antitumor activity in patients with advanced solid tumors[1][4]. Both agents are approved individually for various cancers; their combination aims to enhance efficacy without significant additive toxicity.
