AXL CAR-iNKT is an allogeneic chimeric antigen receptor (CAR) invariant natural killer T (iNKT) cell therapy targeting AXL, a receptor tyrosine kinase overexpressed in various solid tumors, including lung cancer and triple-negative breast cancer (TNBC). Developed by Shanghai Pharma Biotherapeutics and Fudan University, this therapy leverages the unique properties of iNKT cells, which express a highly conserved invariant T-cell receptor (TCR) that recognizes lipid antigens presented by CD1d. Because CD1d is monomorphic across the human population, iNKT cells can be used as an allogeneic, off-the-shelf cell therapy without causing graft-versus-host disease (GvHD). The addition of an AXL-targeting CAR enhances the specificity and cytotoxicity of the iNKT cells against AXL-expressing tumor cells, offering a promising therapeutic strategy for solid tumors.
