Axl-DM1 mAb ADC is a preclinical antibody-drug conjugate (ADC) developed by the National Research Council of Canada. It consists of a conventional monoclonal antibody (mAb) targeting the extracellular domain of the receptor tyrosine kinase AXL (Anexelekto), conjugated to the microtubule-disrupting cytotoxic payload DM1 (emtansine) via a non-cleavable linker. AXL is an oncogenic target upregulated in various cancers, including breast, lung, ovarian, gastric, and prostate cancers, and is associated with epithelial-to-mesenchymal transition, metastasis, and drug resistance. In preclinical studies, Axl-DM1 mAb ADC demonstrated sub-nanomolar cytotoxic potency in vitro against AXL-expressing breast, lung, and ovarian cancer cell lines, and showed potent anti-tumor activity in vivo in ovarian and breast cancer xenograft models.
