Overview

**AXL1717 + gemcitabine + carboplatin** is a combination regimen comprising the small molecule AXL1717 (picropodophyllin, a selective inhibitor of the insulin-like growth factor-1 receptor, IGF-1R), gemcitabine (a nucleoside analog DNA synthesis inhibitor), and carboplatin (a platinum-based DNA crosslinking agent). AXL1717 exerts antitumor effects by inhibiting IGF-1R signaling, thereby blocking tumor cell proliferation and division; it may also arrest mitosis through non-IGF-1R dependent mechanisms. Gemcitabine and carboplatin are established chemotherapeutics used in a range of cancers including NSCLC, bladder cancer, and breast cancer; together, they enhance apoptosis via synergistic inhibition of DNA synthesis and repair. Combination use of these agents is under clinical investigation for synergistic antitumor activity in solid tumors, particularly where IGF-1R is a relevant target[1][2][4][5][7][8].
