AZ59 is a fully human monoclonal antibody that targets the cellular prion protein (PrPC). It was developed to treat Alzheimer's disease by blocking the interaction between amyloid-beta oligomers (Aβo) and PrPC, which serves as a high-affinity receptor for Aβo. By binding to the amino-terminal unstructured domain of PrPC, AZ59 prevents synaptotoxic signaling pathways that lead to synaptic loss and cognitive decline. Preclinical studies in APP/PS1 transgenic mice have shown that systemic administration of AZ59 can rescue memory function and restore synaptic density without requiring the clearance of amyloid plaques. The antibody was developed through a collaboration between AstraZeneca (MedImmune) and Yale University.
