Drug intelligence / Profile preview
AZ7773 is a selective small molecule inhibitor of the NLRP3 (NLR family pyrin domain-containing 3) inflammasome developed by AstraZeneca [2702]. The NLRP3 inflammasome is a multi-protein complex that plays a critical role in the innate immune response by regulating the activation of caspase-1 and the subsequent release of pro-inflammatory cytokines such as IL-1β and IL-18 [2702]. Aberrant activation of NLRP3 is implicated in sterile inflammatory processes, including ischemia-reperfusion injury (IRI) and acute kidney injury (AKI) [2702]. In preclinical porcine models of severe AKI, administration of AZ7773 post-reperfusion has been shown to significantly reduce early kidney injury, improve renal function, and decrease histological injury scores, highlighting its potential therapeutic utility in mitigating AKI [2702].
