Overview

A combination chemotherapy regimen consisting of azacitidine, capecitabine, and oxaliplatin. This regimen was studied in Phase I/II clinical trials for colorectal cancer patients, particularly those with CpG island methylator phenotype (CIMP-high) tumors who were refractory to fluoropyrimidine/oxaliplatin treatment. Azacitidine is a DNA methyltransferase inhibitor that was hypothesized to restore sensitivity to fluoropyrimidine and oxaliplatin chemotherapy through epigenetic modulation. The standard dosing schedule includes subcutaneous azacitidine (75 mg/m²/day on days 1-5), intravenous oxaliplatin (110 mg/m² on day 2), and oral capecitabine (1500 mg/m² divided twice daily for 2 weeks) in a three-week cycle.
