Overview

Azacitidine + carboplatin is an investigational combination therapy pairing azacitidine, a DNA methyltransferase inhibitor, and carboplatin, a platinum-based alkylating agent. Azacitidine induces hypomethylation of DNA, leading to re-expression of silenced genes including those regulating immune response and apoptosis. Carboplatin induces DNA cross-linking, resulting in apoptosis of rapidly dividing cells. The combination is being investigated for potential synergistic effects, such as improved tumor cell killing and enhanced immune activation, particularly in platinum-resistant high-grade serous ovarian cancer. Preclinical and early clinical research indicates that sequential administration may slow tumor growth, reactivate immune pathways (including antigen presentation and immune checkpoint response pathways), and increase rates of apoptosis and cell death compared to monotherapy.
