Overview

Azacitidine + docetaxel is an investigational combination therapy consisting of azacitidine, a DNA methyltransferase inhibitor (hypomethylating agent), and docetaxel, a microtubule inhibitor (taxane class). Azacitidine acts by incorporating into DNA and RNA, leading to hypomethylation of DNA and direct cytotoxicity on abnormal hematopoietic cells. This can reverse gene silencing associated with resistance to chemotherapy. Docetaxel stabilizes microtubules, inhibiting cell division and inducing apoptosis in cancer cells. The combination has been studied primarily in metastatic castration-resistant prostate cancer (mCRPC) patients who have progressed during or after prior docetaxel therapy. Preclinical studies suggest that azacitidine may sensitize tumor cells to the effects of docetaxel by reversing epigenetic changes linked to chemoresistance[1][2][3].
