azacitidine + lenalidomide + grifola frondosa extract is an investigational combination therapy being evaluated by the M.D. Anderson Cancer Center for the treatment of advanced solid tumors and malignancies. This regimen combines the epigenetic modulating effects of azacitidine (a DNA methyltransferase inhibitor) and the immunomodulatory properties of lenalidomide (a cereblon E3 ligase modulator) with a protein-bound beta-glucan polysaccharide extract derived from the Grifola frondosa (maitake) mushroom. The mushroom extract is intended to act as a biologic response modifier, stimulating innate and adaptive immune cells such as macrophages, natural killer (NK) cells, and T cells. The Phase I clinical trial aims to establish the maximum tolerated dose and safety profile of this triplet combination in patients whose cancers are refractory to standard therapies.
