Overview

Azacitidine + tislelizumab is an investigational combination therapy consisting of azacitidine, a DNA methyltransferase inhibitor (epigenetic modulator), and tislelizumab, a monoclonal antibody targeting programmed cell death protein 1 (PD-1). Azacitidine works by inhibiting DNA methylation, leading to reactivation of tumor suppressor genes and increased cancer cell death. Tislelizumab is an immune checkpoint inhibitor that blocks PD-1 on T cells, preventing cancer cells from evading immune detection and destruction. This combination aims to enhance antitumor immunity by both modifying the tumor microenvironment with azacitidine and unleashing immune responses with PD-1 blockade via tislelizumab. The combination is under clinical investigation for various advanced solid tumors and hematologic malignancies.
