Overview

**AzaSite Xtra** is a higher-concentration (2%) topical ophthalmic formulation of azithromycin, a macrolide antibiotic, developed by InSite Vision using their proprietary DuraSite sustained drug delivery technology (polycarbophil-based mucoadhesive polymer). It is designed for treating bacterial conjunctivitis (pink eye) caused by susceptible bacteria, offering prolonged ocular residence time and high tissue concentrations in the cornea, conjunctiva, and eyelids compared to the standard 1% AzaSite. The mechanism involves binding to the 50S ribosomal subunit of bacteria to inhibit protein synthesis. As of available data, it was in development/preclinical evaluation stages, with pharmacokinetic studies in rabbits showing superior drug levels 24 hours post-dose, supporting potential once-daily dosing for 3 days.[3][10][14]
