Overview

AZD-2327 is a highly potent and selective small molecule agonist of the delta opioid receptor (δ-opioid receptor, DOR), developed by AstraZeneca for the treatment of depressive disorders and anxiety disorders. It demonstrated antidepressant and anxiolytic effects in animal models, with a lower risk of tolerance, dependence, and misuse compared to benzodiazepines or other opioid agonists. The drug also acts as an inhibitor of cytochrome P450 CYP3A4. In clinical trials, it showed greater potential for anxiolytic than antidepressant efficacy but was ultimately found ineffective for major depressive disorder in phase 2 studies. Development was discontinued after reaching phase 2 trials[1][2][3][4].
