Overview

AZD-8418 is a small molecule drug developed by AstraZeneca as a positive allosteric modulator of the metabotropic glutamate receptor 2 (mGlu2). It was investigated primarily for its potential to modulate glutamatergic neurotransmission, with the aim of treating neuropsychiatric conditions such as schizophrenia and nicotine dependence. Preclinical studies demonstrated that AZD-8418 attenuates nicotine-taking and nicotine-seeking behavior in animal models, likely by enhancing mGlu2 receptor activity and thereby reducing presynaptic glutamate release. The compound belongs to the class of isoindolones and is administered orally. Clinical development for schizophrenia reached Phase I but was discontinued[1][2][4][5].
