AZD-8848 is a potent, selective small molecule agonist of Toll-like receptor 7 (TLR7), developed as an "antedrug" to minimize systemic exposure by rapid metabolism after local administration. It was initially developed by AstraZeneca for the treatment of asthma and allergic rhinitis, with additional investigation in butyrylcholinesterase deficiency. The drug is designed to stimulate innate immune responses via TLR7 activation, leading to type I interferon production and downregulation of Th2-mediated inflammation. Clinical studies have shown that intranasal or inhaled AZD8848 can reduce allergen responsiveness in patients with allergic asthma by modulating immune pathways while generally being well tolerated[1][3][5][6].
