AZD-9164 is a small molecule drug developed as an inhaled long-acting muscarinic antagonist (LAMA) bronchodilator for the treatment of chronic obstructive pulmonary disease (COPD). It acts primarily as a potent antagonist of the muscarinic acetylcholine receptor M3 (M3 receptor), thereby inhibiting parasympathetic bronchoconstriction and promoting bronchodilation. Preclinical studies demonstrated that AZD-9164 had a longer duration of action than ipratropium in animal models and showed an improved therapeutic index for lung versus systemic effects. Clinical trials reached up to Phase II; however, development was discontinued after observations of transient paradoxical bronchospasm in COPD patients following inhalation[1][2][3][6][7].
