AZD-9684 is a small molecule drug developed as the first member of a new anti-thrombotic class known as carboxypeptidase U inhibitors (CPU inhibitors or TAFI inhibitors). It was designed to enhance endogenous fibrinolysis by inhibiting carboxypeptidase U (also known as thrombin activatable fibrinolysis inhibitor, TAFI, or carboxypeptidase B2), thereby promoting the breakdown of blood clots. The primary indication investigated for AZD-9684 was thrombosis. Despite showing pharmacodynamic effects such as suppression of CPU activity and reduction in fibrin(ogen) deposition in preclinical models, clinical development did not progress beyond Phase 2 and has since been discontinued[1][2][5].
