AZD0022 is an investigational oral small molecule inhibitor developed by AstraZeneca that selectively targets the KRASG12D mutation, a specific alteration in the KRAS gene found in various solid tumors. By inhibiting the function of mutant KRASG12D protein, AZD0022 aims to block downstream signaling pathways involved in tumor cell proliferation and survival. The drug is being developed for use as monotherapy or in combination with other anti-cancer agents such as cetuximab. Its primary indications under investigation include advanced or metastatic solid tumors harboring the KRASG12D mutation, notably pancreatic cancer (including pancreatic ductal carcinoma), colorectal cancer, non-small-cell lung cancer, and adenocarcinoma. As of early 2025, AZD0022 is undergoing Phase I/II clinical trials to assess its safety, tolerability, pharmacokinetics, and preliminary efficacy.
