AZD0284 is a potent, selective, and orally bioavailable small molecule inverse agonist of the human nuclear receptor retinoic acid-related orphan receptor gamma (RORγ), also known as nuclear receptor subfamily 1 group F member 3. RORγ is a master regulator of T-helper 17 (Th17) cell function and differentiation, making it an attractive target for the treatment of Th17-driven autoimmune diseases such as psoriasis. AZD0284 inhibits IL-17A production in primary human Th17 cells and has demonstrated efficacy in preclinical models by reducing IL-17A-producing cells and associated inflammatory markers. The drug was developed by AstraZeneca for potential use in conditions like plaque psoriasis but its clinical development was discontinued after phase I trials[1][2][3][4][5][6].
