AZD0328 is an investigational small molecule that acts as a potent, stereoselective, full agonist of the human α7 nicotinic acetylcholine receptor (α7 nAChR). It exhibits high selectivity for this receptor subtype, with approximately 20-fold greater selectivity over α1β1γδ nAChR and about 1000-fold greater selectivity over other nicotinic receptors and unrelated targets. Preclinical studies demonstrated that AZD0328 enhances cortical dopamine release and improves learning and attentional processes in animal models. In clinical trials, it was evaluated primarily for cognitive impairment associated with schizophrenia and Alzheimer's disease. The drug showed some improvement in operant conditioning and working memory in animal studies but was limited by dose-related nausea in humans. Clinical development reached Phase II for schizophrenia but was terminated due to not meeting target product profiles[1][2][5][7][9].
