Overview

AZD0486 + rituximab is an investigational combination therapy being studied primarily for the treatment of previously untreated follicular lymphoma. AZD0486 is a fully human, bispecific monoclonal antibody (IgG4) that targets both CD19 (on B-cells) and CD3 (on T-cells), functioning as a T-cell engager to promote T cell-mediated cytotoxicity against malignant B-cells. Rituximab is a well-established chimeric monoclonal antibody targeting the CD20 antigen on B-cells, leading to B-cell depletion by complement-dependent cytotoxicity, antibody-dependent cellular cytotoxicity, and induction of apoptosis. The combination seeks to leverage dual mechanisms: direct B-cell targeting (via rituximab) and immune-mediated killing (via T-cell engagement by AZD0486) to improve anti-tumor efficacy in follicular lymphoma compared to standard immunochemotherapy regimens.
