Overview

AZD0754 is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy developed for the treatment of metastatic castration-resistant prostate cancer. The therapy consists of a patient’s own T lymphocytes genetically modified to express a CAR targeting STEAP2, a cell surface antigen highly expressed in prostate cancer. Additionally, these CAR-T cells are "armored" with a dominant-negative transforming growth factor beta type II receptor (dnTGFβRII), which enables them to resist the immunosuppressive effects of TGF-β commonly found in the tumor microenvironment. Preclinical studies have demonstrated that AZD0754 exhibits potent and specific cytotoxicity against STEAP2-expressing tumor cells and maintains antitumor activity even under high levels of TGF-β, supporting its potential as a first-in-class therapy for advanced prostate cancer[1][2][3][5][7].
