Overview

This drug is an **experimental combination therapy** composed of three agents: - **AZD0901**: An antibody-drug conjugate (ADC) targeting Claudin 18.2, a protein highly expressed in gastric, gastroesophageal junction, and pancreatic cancers. AZD0901 consists of a humanized anti-CLDN18.2 IgG1 antibody conjugated via a protease-cleavable linker to the cytotoxic agent monomethyl auristatin E (MMAE). Its mechanism involves specific binding to cancer cells expressing CLDN18.2, internalization, and targeted delivery of MMAE to induce cell death through microtubule disruption, as well as inducing antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC)[3][7]. - **Rilvegostomig (AZD2936)**: A bispecific monoclonal antibody simultaneously targeting PD-1 and TIGIT, major checkpoints in T cell-mediated immune response. By inhibiting both pathways, it is designed to enhance anti-tumor immune activity beyond inhibition of PD-1 alone[6]. - **Fluoropyrimidine**: Refers to a class of cytotoxic chemotherapy agents such as 5-fluorouracil (5-FU) or capecitabine. These are widely used antimetabolites that inhibit DNA synthesis via thymidylate synthase inhibition, leading to impaired tumor cell replication and death[4]. The combination is being studied for treatment of advanced or metastatic solid tumors, especially gastrointestinal malignancies such as gastric, gastroesophageal junction, and pancreatic cancer, as well as HER2-positive and CLDN18.2-expressing tumors. Phase II/III clinical trials are ongoing[4][5][7].
